Trotz ausgereifter Strategien ist die Strukturaufklärung mittels Röntgenbeugung an Kristallen keine Routineaufgabe. Eines der Forschungsziele unserer Arbeitsgruppe ist die Anwendung und Weiterentwicklung der Röntgenstrukturanalyse im Hinblick auf aktuelle Fragestellungen. Hierzu gehört die Untersuchung von supramolekularen Komplexen organischer Moleküle, welche sich nur unter zielgerichteten Kristallisationsbedingungen erhalten lassen. Wir interessieren uns unter anderem für Konformationsänderungen während der Komplexbildung. Ein einfaches Beispiel hierfür sind Acetylhydrazone, die in Abhängigkeit von der Größe der Substituenten bevorzugt Dimere oder Polymere bilden. Wir haben diese Untersuchungen erfolgreich auf Systeme ausgedehnt, die aus verschiedenen Komponenten bestehen, und uns seit kurzem Wirkstoff/Rezeptor-Komplexen zugewandt [1].
Häufig kommt es zu erheblichen Problem bei der Lösung des kristallographischen Phasenproblems. Das von uns entwickelte Programm PATSEE hat nicht nur viele Problemstrukturen gelöst [2], sondern ist auch erfolgreich eingesetzt worden, um Kristallstrukturen aus Pulverdaten zu bestimmen, wenn kein geeigneter Einkristall zur Verfügung steht. Für den Einsatz von PATSEE müssen allerdings wesentliche Teile der dreidimensionalen Molekülstruktur bekannt sein.
Zu diesem Zweck nutzen wir das in unserer Arbeitsgruppe entwickelte Kraftfeldprogramm MOMO. Es basiert auf dem PIMM85-Kraftfeld und beinhaltet z. B. eine Potentialfunktion für Wasserstoffbrücken, Modelle zur Berechnung atomarer Punktladungen und die Behandlung elektrostatischer Wechselwirkungen durch Multipolmomente. MOMO wurde unter dem Gesichtspunkt entwickelt, eine Umgebung für spezielle problemorientierte Erweiterungen bereitzustellen. Der Programmteil SupraMol ermöglicht beispielsweise die Berechnung supramolekularer Komplexe, die aus Molekülen mit mehreren Torsionsfreiheitsgraden gebildet sein können.
Immer wieder war es notwendig, das Programm an neue Rechnergenerationen, Betriebssysteme und Programmierwerkzeuge anzupassen. Die aktuelle Version ist in den Programmiersprachen Python und C geschrieben, weshalb wir es nun als PyMOMO bezeichnen (www.pymomo.de). Aktuell haben wir PyMOMO mittels Referenzdaten, die aus quantenmechanischen Rechnungen stammen, erfolgreich reparametrisiert [3]. Außerdem arbeiten wir an der Erfassung von C-H∙∙∙X-Wasserstoffbrücken und der Implementierung eines modernen und effizienten Solvatationsmodells.
